AFAIK, using TWRP for a full backup requires an unlocked bootloader [1]. Normally the bootloader is locked, because an unlocked bootloader is a security risk in case the phone falls into the wrong hands. For the same reason, on most phones unlocking the bootloader (via an unlocking code from the manufacturer or third party) will do a factory reset, deleting all the very programs and data you're trying to backup. Catch-22. So how did you get to flash TWRP on your Nexus without these problems?
